Blueberries (Vaccinium), belonging to the Ericaceae family, are propagated using strategies such as micropropagation to optimize clonal production. From this perspective, this study aimed to improve the acclimatization process of blueberry plants by evaluating the effect of different flask sealing methods during in vitro cultivation. Three cultivars—Bluegem, Climax, and Woodard—were used in the experiment. Explants were induced to root in a medium composed of water, 5.5 g L−1 agar, and 20 mg L−1 indole-3-butyric acid (IBA), then transferred to flasks containing Woody Plant Medium (WPM) supplemented with activated charcoal. The cultures were maintained in a growth chamber for 30 days under three sealing conditions: standard lids, plastic film, and lids with filters. Following this period, explants were transplanted to polyethylene trays containing a commercial substrate and acclimatized in a greenhouse equipped with an intermittent microspray system for 60 days. The experiment included morphometric, histological, survival, and photosynthetic pigment analyses. Results varied among cultivars. No significant differences in morphometric parameters were observed among the three cultivars during the in vitro proliferation stage. However, differences emerged in survival rates and in morphometric and pigment-related parameters at the end of acclimatization. The use of filter lids improved the micropropagation and acclimatization efficiency of the Climax cultivar. Additionally, filter-sealed flasks enhanced survival rates in Bluegem, whereas no notable improvements were observed for the Woodard cultivar. These findings highlight the importance of optimizing flask sealing systems in tissue culture to develop more efficient acclimatization protocols, thereby increasing the quantity and quality of plantlets obtained. The use of filter-sealed flasks improved the acclimatization efficiency of the Climax blueberry cultivar and the survival rates of the Bluegem cultivar.

(en) Abstract: Whether gypsum application changes mineral composition of Eucalyptus urograndis bark and wood along a tree height gradient is uncertain. Therefore, an experiment was established to examine three rates of broadcasted gypsum (1.2, 4.8, and 9.6 Mg ha-1; control); applied before planting. Three years after application, discs were collected to obtain bark and wood chemical composition from the tree base and at 25, 50, 75, and 100% of total height. Results shows that calcium (Ca), iron (Fe), aluminum (Al), and manganese (Mn) can accumulate in wood at the tree base, while potassium (K), sulfur (S), and phosphorus (P) tend to accumulate in the upper portions of trees. Gypsum application increased concentrations of Ca, S, and P in wood; Ca, S, P and copper (Cu) in bark; and decreased Mn in bark and wood. Findings suggest that high gypsum rates increased exportation of Ca, S and P by wood. Additionally, to accurately estimate nutrient exportation it is important to sample at multiple height positions along the tree trunk.

(pt) This study evaluated the morphophysiological characteristics of sugarcane buds from different stalk positions of the Havaianinha variety under varying air temperatures, aiming to understand biotic and abiotic factors affecting seedling production via the pre-sprouted seedling (PSS) system. Two experiments were conducted at the Micropropagation Laboratory of the Federal University of Paraná, using a completely randomized design (CRD) in a 4 × 3 factorial scheme (four temperature levels: 15, 20, 25, and 30 ºC; three bud positions: apical, median, and basal) with three replications. Each experimental unit contained 20 mini setts. The sprouting speed index (SSI), sprouting percentage (SP), and sprouting height (SH) were evaluated. Data were analyzed using ANOVA, Pearson correlation, Scott-Knott test (5%), and regression analysis for temperature. SSI, SP, and SH were influenced by mini sett age, temperature, and bud position. The highest values for all parameters occurred at 30 ºC, using apical buds from 18-month-old culms or buds from any position on 6-month-old culms. Temperatures below 20 ºC negatively affected all variables. Positive correlations among SSI, SP, and SH were observed. For optimal PSS seedling production, younger culms should be used to maximize bud utilization.
